Truncated or soluble EGFR (sEGFR), sErbB2 and sErb3 have been identified in a variety of species including the human. sEGFR is present in normal human serum and urine. sEGFR has also been implicated in multiple cancers. Research suggests sEGFR plays a biological role in both normal and cancerous cells. However, the overall function of sEGFR remains unclear. Therefore, the specific aim of this dissertation was to investigate biological roles of sEGFR in the mammary gland, specifically its ability to act as a signaling protein and elicit physiologic effects in mammary epithelial cells. Soluble receptor binding assays conducted with purified sEGFR confirmed specific binding, with high affinity, to epidermal growth factor (EGF). Binding assays with intact mammary epithelial cells also result in sEGFR binding. This binding can be inhibited by 50% with EGF competition. The protein phosphorylation states of multiple proteins were increased in response to sEGFR in normal mammary epithelial cells (NMuMG). This increase was not observed in mouse fibroblast (B82L) cells lacking EGF. NMuMG cells treated with sEGFR using western blot analysis showed an increase in tyrosine specific phosphorylation of Cytokeratins 8 (CK8), 18 (CK18), 19 (CK19) and gamma-actin in comparison to control and mutated sEGFR (mut-sEGFR) treated cells. 2D-PAGE coupled with MALDI-MS yielded protein expression changes in NMuMG cells in response to sEGFR. CK8, Phospholipase C-alpha and tubulin alpha-6 all increased in response to treatment. CK8 resolved as three independent spots, most likely due to different phosphorylation states of the protein. sEGFR increased whole cell protein expression of CK8 significantly upon 1hr and through 24hr treatments.
